三溴化硼,无色透明液体,Lewis酸,溶于二氯甲烷、四氯化碳、正戊烷,二硫化碳等溶剂,是一种非常高效的温和的脱甲基试剂【BBr3裂解醚反应】。当底物分子中杂原子数很多时,应增加相应BBr3量。使用BBr3有一个最大的缺点是BBr3对空气敏感,使用时会冒出大量气雾。脱甲基反应时,常用其1~3M的二氯甲烷溶液(4~10个当量),在-78 ℃下加入,室温下搅拌反应。在同时存在甲基醚和苄基醚时,控制在低温下反应,可以选择性脱苄基醚保护。

常用后处理方法一:反应完全后,低温下加水淬灭反应,中和,萃取得到产物。加水后处理时会生成硼酸,如果产物难以从水中萃出,此时最好使用其它的方法,否则后处理艰难并导致收率下降。常用后处理方法二:将反应液和适量甲醇分别冷却至-78℃,将反应液缓慢加入冷却的甲醇中淬灭反应,生成的硼酸三甲酯可以减压蒸除。此方法一定要控制好内温,防止局部温度过高起火(否则你会看到蓝色的小火苗!)常用后处理方法三:如果底物稳定性好,可以参考三氯氧磷后处理方法,直接旋蒸旋掉剩余的三溴化硼,然后再用甲醇淬灭,避免生成大量的酸气。另外一些溶解度较差底物进行脱甲基时,当量的三溴化硼很难完全反应,有时直接用BBr3/DCM(1:1)进行反应,脱除效果可能会不错。这种情况,就不适合用甲醇淬灭反应,先旋掉大量的三溴化硼后再进行后处理,操作会简化很多。常见的副反应:1、底物上含有易离去基团时,可能会和溴进行取代置换,如,吡啶吡嗪等氮杂环2位氯,脂肪卤代烃,醇羟基等;2、三溴化硼脱甲基时,酯可能会发裂解生成羧酸,遇到这种情况,不要急于后处理,如果是甲酯的话,直接将反应液降温至-78℃,缓慢加入甲醇,升至室温下搅拌,可以重新酯化。
