个人简历

1963年9月出生,安徽人,博士毕业于南京大学生物化学与分子生物学专业,1996年12月至2001年8月先后在美国罗彻斯特大学医学中心和加拿大多伦多大学附属病童医院(SickKids)接受博士后训练。2001年9月作为教育部“长江学者”特聘教授,受聘为东南大学基础医学院遗传学研究中心主任,担任博士生导师;领衔申报东南大学第一个生命科学领域博士授权点(遗传学)获准;2003年获准建设“发育与疾病相关基因”教育部重点实验室,担任实验室主任;2004年4月至2009年11月担任东南大学基础医学院院长,2009年12月至2019年12月担任东南大学生命科学研究院院长,期间完成了东南大学生物学一级学科硕士点、博士点、博士后流动站的建设、协助基础医学博士点的获准;2019年领衔论证成立生命科学与技术学院,获得东南大学批准,谢维任学院学术委员会主任、东南大学首席教授。

谢维教授先后获得国家自然科学基金(面上、重点)、中-加合作项目、国家杰出青年科学基金(2003年)、美国NIH基金(RO3)、国家“973”重大基础研究项目、国家科技创新2030-“脑科学与类脑研究”(“国家脑计划”)重大研究项目(首席科学家)等资助。长期从事脑发育的遗传控制、跨细胞与跨系统信号调节机制和抗体工程靶向药的研究。发现了自闭症关联基因参与调控突触、环路发育;解析了调控社交行为的多条神经环路等;指导创业团队研发抗体靶向药,已获得临床试验许可证6张(中、美),其中两款药物分别在临床I、II期实验。

小鼠早期器官发生过程中完整胚胎的数字化重建

亮点  在早期器官发生过程中,以单细胞分辨率重建完整数字胚胎, 构建中胚层和内胚层谱系的时空图谱和信号通路图谱, 识别胚-胚外界面处的原基决定区 (PDZ), 表征参与器官原基形成的跨胚层信号

总结  早期器官发生是胚胎发育的关键阶段,其特点是广泛的细胞命运决定以启动器官形成,但也极易出现发育缺陷。在此,我们对来自 6 个 E7.5-E8.0 胚胎的 285 个连续切片进行了分析,以单细胞分辨率构建了早期器官发生过程中的完整时空转录组和信号通路图谱。通过开发 SEU-3D,我们重建了数字胚胎,从而能够在天然的空间环境中研究区域化的基因表达。我们建立了一种基于空间信息的基因-细胞共嵌入方法,系统地构建了内胚层和中胚层衍生物的空间图谱,并阐明了跨胚层和细胞类型的信号网络。此外,我们还表征了在胚胎发育第7.75天(E7.75)沿胚胎-胚外界面形成的原基决定区(PDZ),并揭示了协调的信号通讯促进了心脏原基的形成。总而言之,这种高分辨率的“数字胚胎”为早期器官发生提供了重要的见解,并为研究发育和疾病提供了一个独特的空间平台。
